ABSTRACT

ObjectiveTo develop a quality control method for the simultaneous determination of multiple active components in Nymphaeae Flos aiming at the problems of the single index for quality control and the relatively low overall quality control level. MethodUltra performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS)was used to identify and select the index components for quality control with the mobile phase of 0.1% formic acid aqueous solution(A)-acetonitrile(B)for gradient elution (0-2 min, 3%-8%B; 2-4 min, 8%-10%B; 4-13 min, 10%-15%B; 13-19 min, 15%-20%B; 19-26 min, 20%-45%B) at a flow rate of 0.4 mL·min-1, detection wavelength of 350 nm, electrospray ionization(ESI), negative ion scanning mode, ion source temperature of 120 ℃, scanning range of m/z 100-1 200, transmit collision energy of 6 eV for low-energy scanning and 25-50 eV for high-energy scanning. High performance liquid chromatography(HPLC)was used to establish the quality control method for the simultaneous determination of multi-index components with the mobile phase of 0.2% phosphoric acid aqueous solution(A)-acetonitrile(B) for gradient elution(0-30 min, 12%-15%B; 30-60 min, 15%-22%B; 60-90 min, 22%-40%B)and detection wavelength of 350 nm. The preparation method of the test solution for content determination was refluxing extraction for 60 min with 80 times the amount of 70% methanol. ResultBy comparing the retention time, ultraviolet absorption characteristics, MS and MS/MS spectrometric signals in the samples with the reference substances, 8 active components with high contents, including brevifolincarboxylic acid, ellagic acid, rutin, nicotiflorin, astragalin, quercetin, quercetin-3-methylether and kaempferol, were identified qualitatively from Nymphaeae Flos, which were selected as the index components for quality control. Under the established HPLC conditions, the above 8 components could be well separated(resolution>1.5), and showed good linearity(r=0.999 9)between the concentration ranges of 1.99-99.6, 1.76-176, 1.52-75.8, 3.60-180, 0.964-96.4, 1.18-118, 1.94-96.8, 1.04-104 mg·L-1 and the peak areas, respectively. The detection limits of them were 10-49 μg·L-1, and the limits of quantitation were 34-164 μg·L-1. The average recoveries were 97.12%-103.1% with the relative standard deviations (RSDs) were 1.1%-2.2%. ConclusionA quality control method for simultaneous determination of the multiple active components in Nymphaeae Flos have been developed, which is simple, accurate and reproducible, and it can provide a scientific basis for the formulation of quality standard of this herb and lay a research foundation for the transformation of Uygur hospital preparations containing Nymphaeae Flos into new drugs.

ABSTRACT

ObjectiveTo explore the antidepressant effect of Sophora flavescens seed extract and its molecular mechanism. MethodA mouse depression model was established by intraperitoneal injection of lipopolysaccharide(LPS), and normal group, model group, fluoxetine group(2.5 mg·kg-1), and S. flavescens seed low, medium and high dose groups(200, 400, 800 mg·kg-1) were set up for 7 d of consecutive gavage. Then the antidepressant effect of S. flavescens seed extract was evaluated by using open field test, elevated plus maze test and forced swimming test. Pathological morphological changes in the hippocampal tissue was observed by hematoxylin-eosin(HE) staining. Protein expression levels of G1/S-specific cyclin D1(Cyclin D1), Wnt1, β-catenin and phosphorylated glycogen synthase kinase-3β(p-GSK-3β) in mouse brain tissues were detected by Western blot. Hippocampal cell apoptosis was detected by terminal deoxynucleotidyl transferase mediated deoxyuridine triphosphate(dUTP) nick end labeling(TUNEL). ResultThe results of mouse behavioral experiments showed that compared with the normal group, the speed of movement in the open field and the distance of movement in the central area of the open field, and the time spent on the open arms of the elevated plus maze were significantly reduced in the model group(P<0.01), while immobility time in the forced swimming test was significantly increased(P<0.05). Compared with the model group, the S. flavescens seed medium and high dose groups had increased speed of movement in the open field test and time spent on the open arms of the elevated plus maze test(P<0.05, P<0.01), and decreased immobility time in the forced swimming test(P<0.05), the distance of movement in the central area of the open field test increased in the high dose group(P<0.05). HE staining results showed that compared with the normal group, the hippocampal neuron structure of mice in the model group was damaged. Compared with the model group, after treatment of S. flavescens seed extract, the pathological state of the mouse hippocampal neuron structure was alleviated, and the neurons increased, were neatly arranged, and the cytoplasm was clear. Western blot results showed that the protein expression levels of Wnt1 and β-catenin in mouse brain tissue were significantly decreased(P<0.01), while the protein expression levels of Cyclin D1 and p-GSK-3β were significantly increased(P<0.01) after LPS injection. Compared with the model group, protein expression levels of Wnt1 and β-catenin in brain tissue of S. flavescens seed medium and high dose groups were significantly increased(P<0.01), while the protein expression levels of Cyclin D1 and p-GSK-3β were significantly decreased(P<0.01). TUNEL staining results showed that the hippocampal cell apoptosis rate in the model group was significantly increased compared with that of the normal group(P<0.01), while the hippocampal cell apoptosis rate in the S. flavescens seed medium and high dose groups was significantly decreased compared with that of the model group(P<0.01). ConclusionS. flavescens seed extract can effectively improve the severity of depression in LPS-induced depressed mice, and its molecular mechanism is related to the regulation of neuroinflammation and hippocampal neuronal apoptosis mediated by Wnt/β-catenin signaling pathway.

ABSTRACT

Objective@#To observe the joint effect of school and family tobacco control on middle school students, and to explore its effectiveness in reducing second-hand smoke exposure in middle school students’ families, as so asto provide a new way of thinking for controlling smoking among middle school sudents.@*Methods@#A questionnaire survey was conducted on the study of 2 125 primary and high school students in four middle schools in the Dongcheng District of Beijing by means of a stratified group sampling method. Personal information of students, tobacco control environment around the family, school and surrounding ares, as well as the knowledge, attitude and behavior of students related to tobacco control was collected.@*Results@#The exposure rate of second-hand smoke in the families of middle and high school students in Dongcheng District of Beijing was 33.41%. Smoking by at least one parent was positively correlated with second-hand smoke exposure in the student’s home (OR=22.88, P<0.05). Among the students who saw the "clear no-smoking label" on campus, the exposure rate (4.92%) when the smoking restriction was set on the home was lower than that of those who did not have a regulation in the home (58.94%) (χ2=452.57, P<0.01). The results of Logistic regression showed that there was a combination of family smoking restrictions and second-hand smoke exposure in middle and high school students who had seen the smoking ban on campus (OR=0.08) and had the school’s tobacco control education (OR=0.08).@*Conclusion@#The joint tobacco control measures between school and family can effectively reduce the exposure rate of second-hand smoke in middle school students’ families, which is better than the effect of tobacco control alone.

ABSTRACT

To investigate the relationship between Snail and renal tubular epithelial-mesenchymal transition (EMT) or tubulointerstitial fibrosis (TIF) at tissue and cellular levels and to observe the changes of miRNA profile after transfecting Snail gene into human renal tubular epithelial cells (HK-2),to further elucidate the importance of miRNA in the pathogenesis of renal fibrosis.Methods The expression of Snail and EMT-related proteins vimentin,SMA,E-cadherin was detected by immunohistochemistry in renal tissues of 40 patients with IgA nephropathy.The expression of Snail,E-cadherin and SMA in normal control group,empty transfection group and Snail gene transfection group was detected by Western blot and RT-PCR.Furthermore,differentially expressed miRNAs were screened by gene chip.Results By immunohistochemistry,Snail expression was positively correlated with vimentin and SMA,negatively correlated with E-cadherin in IgA nephropathy.The higher degree of the TIF,the stronger the expression of Snail.Compared with the control group,the expression of Snail,vimentin and SMA in the snail transfected group increased.However,E-cadherin decreased at gene and protein level by the RT-PCR and Western blot (P < 0.05).The difference was statistically significant.Five distinctly different miRNAs were screened by gene chip after Snail gene was transfected into HK-2 cells,and then 5 026 potential target genes were predicted.Conclusion Snail expression is closely related with renal tubular epithelial mesenchymal transition and tubulointerstitial fibrosis,and it may be used as a new target in EMT prevention.Differentially expressed miRNAs may be involved in the development of EMT and TIF.

ABSTRACT

To investigate the epidemic characteristics and prevention strategies of main infectious diseases. From 2010 to 2012, 23 notifiable diseases were reported in Yantai with average reported cases of 17,376. The morbidity was the highest in 2012 and the lowest in 2011. The accumulated death toll was 101 with the highest death rate in 2011 and the lowest in 2012. Major class A and B infectious diseases included viral hepatitis, phthisis, syphilis, clap and dysentery. The major class C infectious disease was hand-foot-and-mouth disease every year. The distribution of transmission routes of HIV/AIDS among patients with HIV and AIDS in Yantai were investigated. The overall condition of phthisis in Yantai showed stable with slight decrease. The occurrence rate of phthisis every season had fluctuation with higher rate in middle two seasons and lower rate at the beginning and end. The major type of viral hepatitis was still hepatitis B. Patients with hand-foot-and-mouth disease were children under 5 years old accounting for 90.56%, mainly reported in May, June, July and August. At present, the condition of plague prevention for infectious diseases in Yantai is not optimistic with both new and old infectious diseases

ABSTRACT

Objective To evaluate the prophylactic effect of abdominal breathing exercises on radiation pneumonitis in patients with chest malignant tumors.Methods 100 patients with chest malignant tumors were given radiotherapy and divided with random digit table into the exercise group and the control group with 50 patients in each group.The incidence rate of radiation pneumonitis in both groups was compared.The prophylactic effect of abdominal breathing exercises on radiation pneumonitis was evaluated.Results The incidence of radiation pneumonitis in abdominal breathing exercise group was significantly reduced [34% (17/50) vs.56% (28/50),Z=2.397,P<0.05],the incidences of grade Ⅰ [22% (11/50) vs.30% (15/50)] and grade Ⅱ [12% (6/50) vs.18% (9/50)] radiation pneumonitis decreased,and no grade Ⅲ and Ⅳ radiation pneumonitis occurred.Conclusions The incidence of radiation pneumonitis in patients with chest malignant tumors treated with radiation therapy may be decreased by abdominal breathing exercises,which may be widely applied in patients treated with radiation therapy.

ABSTRACT

Objective To assess the efficacy and safety of lamotrigine for absence seizures in children and adolescents . Methods Databases of PubMed ,the Cochrane Library ,EMbase ,CENTRAL ,VIP ,WanFang ,CBM and CNKI were electron-ically searched till August ,2014 for clinical trials on lamotrigine for absence seizures in children and adolescents .All literature were screened by two reviewers independently according to the inclusion and exclusion criteria .The data was extracted ,and the methodological quality was assessed .Then ,meta-analysis was performed using RevMan 5 .2 .Results Seven trials were in-cluded involving a total of 721 patients .The results of methodological qualities were two studies rated as A-class ,three studies rated as B-class and two studies rated as C-class .Meta-analysis results showed that the efficacy of lamotrigine monotherapy for absence seizure in children and adolescents was better than placebo ,but efficacy of lamotrigine was lower than valproic acid and ethosuximide .The adverse reaction rates of lamotrigine were with no significant difference compared with valproic acid and et-hosuximide .Conclusion Lamotrigine monotherapy was effective for absence seizures in children and adolescents and was well tolerated .Lamotrigine was a good choice for patients that are intolerable to valproic acid or ethosuximide .

ABSTRACT

Objective To study the effects of tyrosine kinase receptor B-brain-derived neurotrophic factor (TrkB-BDNF) signal pathway on the secretion of vascular endothelial growth factor (VEGF) and matrix metalloproteinases-9(MMP-9) of neuroblastoma.Methods We used all-trans retinoic acid (ATRA) to induce the high expression of TrkB in the SH-SY5Y cell line,and then added the ectogenid BDNF to activate the TrkB-BDNF and its three downstream signal pathways.TrkB-BDNF signal pathway was inhibited by specific tyrosine kinase inhibitor K252a.The three downstream signal pathway was respectively inhibited by LY294002 (the phosphatidylinositol 3-hydroxy kinase (PI3 K) pathway inhibitor)、U73122 (the phospholipase C pathway inhibitor) 、U0126(the mitogen activated protein kinase pathway inhibitor).Enzyme linked immunosorbent assay was used to detect the concentration of VEGF and MMP-9 protein in the SY5Y cell culture supernatants.Results VEGF [(485.89 ± 109.99) pg/ml] and MMP-9 [(15.73 ± 1.72) pg/ml] protein levels in neuroblastoma cells cultured in serum-free media in the group of ATRA + BDNF were significantly higher than that of the control group and ATRA alone group(P <0.05).VEGF [(272.42 ±86.33) pg/ml]and MMP-9 [(5.25 ± 1.44) pg/ml] protein levels in the group of ATRA + BDNF + K252a were significantly lower than those of the ATRA + BDNF group(P < 0.05) and had no significant difference compared with the control group and the ATRA alone group(P >0.05).VEGF [(314.12 ±24.68) pg/ml] and MMP-9 [(4.91 ± 1.08) pg/ml] protein levels in the group of ATRA + BDNF + LY294002 were significantly lower than those of the ATRA + BDNF group(P < 0.05) and had no significant difference compared with the control group and the ATRA alone group(P >0.05).VEGF [(444.08 ±64.49) pg/ml] and MMP-9 [(13.28 ±3.38) pg/ml] protein levels in neuroblastoma cells cultured in serum-free media in the group of ATRA +BDNF + U73122 had no significant difference compared with the ATRA + BDNF group(P > 0.05).VEGF [(429.97 ± 19.95) pg/ml] and MMP-9 [(13.96 ± 4.45) pg/ml] protein levels in neuroblastoma cells cultured in serum-free media in the group of ATRA + BDNF + U0126 had no significant difference compared with the ATRA + BDNF group(P > 0.05).Conclusion Activation of TrkB-BDNF signal pathway can increase the synthesis and secretion of VEGF and MMP-9 in human neuroblastoma cells.TrkB-BDNF signal pathway may be through activating its downstream PI3K pathway to increase the synthesis and secretion of VEGF and MMP-9 in human neuroblastoma cells.The synthesis and secretion of VEGF and MMP-9 can be inhibited by blocking the TrkB-BDNF signal pathway with K252a or blocking its downstream signal pathway PI3 K with LY294002.

ABSTRACT

ObjectiveTo understand the behavioral characteristics,strength on suicide ideation and mental disorder diagnosis among suicide attempers,so as to provide consultation for crisis intervention of suicide.Methods232 suicide attempters from emergency of 5 general hospital in south-west of Shandong province were enrolled.They were assessed using questionnaires on attempted suicide behaviors and scale on the strength of suicide ideation,and were diagnosed using the Structured Clinical Interview for DSM-Ⅳ Disorders axis Ⅰ for patient (SCID/I-P).Results( 1 ) In the total of 232 cases,female was more than male with the gender ratio of 2.87 (female vs male).The highest proportion rate (56.9%) existed between 16 years old to 35 years old.Oral application pesticide was the most commom suicide pattern (83.2%),91.4% of the suicide attempt occurred in the their homes.The top three reasons leading to suicide were family contrary (61.2%),depressive mood (15.1%)and fail in love (9.5%).(2) In all cases,83.6% did not think if they were found,84.1% did not adopted any precaution measures,over 60.0% did not tell anyone their attempted plans,and 90% had no dying structions and /or dying testaments before their attempted behaviors.At the moment of attemp behaviors,62.9% did not contact with someone.The 60.0% of all the cases had wanted strongly to die,otherwise 7.3% had wanted to manipulate others.(3) The diagnosis rate of mental disorder on DSM-Ⅳ was 45.3%,in which the mood disorder was most.ConclusionThere are no changes on the behavioral characteristics among the suicide attempters in the south-west rural area of Shandong province.Their strength of suicide ideation are moderate,and a certain proportion of suicide attempters may be impulsive suicide attempters.There is more relationship between mental disorder and suicide attempt.

ABSTRACT

On the basis of analysis and summary of current application of reference extracts of traditional Chinese medicine, reference extracts for quality assessment of traditional Chinese medicines are divided into three classes by purpose: 1. herbal or volatile oil reference extracts for TLC identification of original herbal medicines or patent traditional Chinese medicines containing original herbal medicines. 2. component reference extracts for identification of certain components or positioning of chromatographic peaks. 3. reference extracts marked with component content to be determined for quantitative analysis on samples. For the third kind of reference extracts, akebiae saponin was taken as an example for preparation thought and methods in such aspects as preparation process, component identification and content analysis.

ABSTRACT

The present study was conducted to develop new inhibitors of pancreatic lipase and alpha-glucosidase from Chinese dietary herbs. Sixty-three dietary herbs from 39 taxonomic families were selected and extracted with aqueous ethanol or water. The extracts were then tested with in vitro enzyme assays for their ability to inhibit pancreatic lipase and alpha-glucosidase activities. Orlistat and acarbose were used as two positive controls. The extracts of Nelumbo nucifera, Curcuma longa, Piper longum and Morus alba showed strong pancreatic lipase inhibitory effects with IC50 at (28.00 +/- 5.51), (5.24 +/- 0.51), (14.76 +/- 2.58), (4.78 +/- 0.58), (3.41 +/- 0.67) mg x L(-1), respectively. These extracts also showed potent alpha-glucosidase inhibitory activities with IC50 at (1.98 +/- 0.13), (0. 18 + 0.007), (0.71 +/- 0.08), (0.077 +/- 0.005), (0.089 +/- 0.006) g x L(-1), respectively. The results provide useful information for developing new drugs or natural health products for hyperlipidemia and hypoglycemia from Chinese dietary herbs.

ABSTRACT

Eight compounds were isolated from Bufonis periostracum by repeated column chromatography on silica gel, ODS and Sephadex LH-20 and their structures were characterized as palmitatic acid cholesteryl ester (1), cholesterol (2), 5alpha, 8alpha-epidioxycholesta-6-en-3beta-ol (3), cholest-5-en-3beta, 7beta-diol (4), cholest-7-en-3beta, 5alpha, 6beta-triol (5), 3-octaddecyloxy-1, 2-propanediol (6), isisamide (7) and bufothionine (8) on the base of spectral analysis. Compounds 1-8 were isolated from Bufonis periostracum for the first time and compounds 3, 5, 6, 7 were obtained from Bufo bufo gargarizans and Bufo genus for the first time. The bioassays showed all tested samples displayed no antitumor activity against the cell lines such as A549, BeL 7402, HGC-27 and HL-60, except the control compound bufalin.

ABSTRACT

Objective To evaluate the value of haptoglobin (HPT),α1-acid glycoprotein (AAG)and C-reactive protein (CRP) in the differential diagnosis of bacterial and viral infection in children. Methods Serum CRP, AAG and HPT levels were measured in 64 children with bacterial infection and 64 children with viral infection during acute stage. These indicators were also measured in 31 normal control children. The sensitivity ,specificity ,positive and negative predictive values, Youden's index for HPT,AAG and CRP were calculated. These values were compared among three groups. Their early diagnostic value on bacterial infection and virus infection was evaluated. Results (1) These markers were considered positive if HP≥2 g/L,AAG≥2 g/L,CRP≥10 mg/L. (2) The serum CRP [(2.70 ±0.99) g/L vs (1.42 ±0.75) g/L] ,AAG [(0.91 +0.38) g/L vs (0.30+0.08) g/L] and HPT [(6.40 ±0.89) mg/L vs (1.38 +0.80) mg/L]levels were significantly higher in children with bacterial infection compared with those with viral infection during the acute stage (P < 0. 05,P < 0. 01). The serum CRP, AAG and HPT levels were significantly higher in children with bacterial infection compared with normal control children during the acute stage (P < 0. 05);while the levels of these indicators showed no differences between viral infection children and normal control children (P > 0. 05). Conclusion The measurement of serum CRP, AAG and HPT levels has clinical value in the differential diagnosis of bacterial and viral infection in children. Among these indicators, CRP has the highest specificity,positive predictive,negative predictive values and Youden's index.

ObjectiveTo compare the clinical and social-psychological characteristics of patients with and without mental disorder.MethodsBase the diagnosis of mental disorder,232 suicide attempters were divided into two groups:suicide attempter with mental disorder ( 105 cases) and suicide attempter without mental disorder (127 cases).The Structured Clinical Interview for DSM-Ⅳ,the Strength of Suicide Ideation,Health Questionnaire of WHO,Questionnaire of Life Measure (QLM),Beck Depression Inventory (BDI) and Beck Anxiety Inventory (BAI),Barratt Impulsiveness Scale (BIS) and Buss Aggressiveness Scale (BAS),Social Support Rate Scale and Questionnaire of Coping Style (QCS) were conducted.ResultsThe 105 suicide attempters with a mental disorder were older than the 127 suicide attempter without a mental disorder ( 37.9 ± 14.5 vs 32.9 ± 13.3,t =2.710,P =0.007 ),and the percentage of female was lower in the former than in the later(65.7% vs 81.1%,x2 =7.099,P =0.008 ).In the total of 232 cases,female was more than male with the gender ratio of 2.87 ( female vs male).The strength level of suicide ideation was higher ( 7.1 ± 2.8 vs 4.8 ± 2.3,t =6.498,P =0.000).The scores of health questionnaire of WHO,BDI and BAI were higher in the suicide attempters with a mental disorder than those without a mental disorder,otherwise the scores of QLM,BIS and its three sub-scales were higher.Among QCS,the sub-score of Self-blaming was higher,and the sub-scores of Question solvation and Asking for help were lower.ConclusionThere are distinguished differences on the sociodemographic and psychological characteristics among the suicide attempters with and without a mental disorder.Mental diseases play an important role in the development of suicide attempter with a mental disorder.

ABSTRACT

Objective To explore the roles on psychosocial factors of suicide behavior in the suicide attempters.Methods132 suicide attempters and 132 matched controls treated in the emergency department were assessed using certain psychosocial questionnaires,such as Health Questionnaire of WHO,Questionnaire of Life Measure ( QLM),Beck Depression Inventory (BDI) and Beck Anxiety Inventory ( BAI),Barratt Impulsiveness Scale (BIS) and Buss Aggressiveness Scale ( BAS),Social Surpport Rate Scale and Questionnaire of Coping Style (QCS).Results( 1 ) The results of single-factor analysis showed that the scores of Health Questionnaire of WHO and QL were lower in the suicide attempter group than those in the control group( suicide attempter group:17.07±3.07 and 9.76 ±7.11; control group:13.42 ±3.31 and 17.08 ±3.81,all P<0.05),otherwise the scores of BDI,BAI,BIS and BAS were higher( suicide attempter group:15.22 ± 11.22,27.87 ± 7.24,87.67 ±20.08 and 73.74 ± 19.61 ; control group:6.87 ± 6.36,26.22 ± 5.44,73.34 ± 14.54 and 63.78 ± 16.44,all P < 0.05 ).Among QCS,the sub-score of self-blaming was higher( 3.42 ± 2.44 VS 2.35 ± 2.04,P < 0.05 ),otherwise the sub-scores of question solvation,asking for help and rationalization were lower( all P < 0.05 ).( 2 ) The result of multi-factors analysis showed that gender,mean education times,the scores of QL,HQ,BDI,and BIS,sub-score of rationalization,and so on,entried the Logistic aggressive equaltion.Conclusion The status of health,quality of life,depressive and anxious mood,impulsive and aggressive personality,and certain coping style play important role.

ABSTRACT

<p><b>OBJECTIVE</b>To remove plumbum ion from the extract of Lonicera japonica by gamma-mercaptopropyl-modified silica gel (MPS).</p><p><b>METHOD</b>The reference plumbum ion solution was added into the extract of L. japonica as the tested samples. The static and dynamic adsorptions were investigated to evaluate the influent factors on the removal of plumbum ion in the extract. The solid content and HPLC profiles of the extract were determined to evaluate the process changes.</p><p><b>RESULT</b>MPS showed excellent adsorption charateristics to remove plumbum ion from the extract of L. japonica, with fast adsorption rate and more than 80% removal efficiency. The optimum parameters were as follows: 6 times sample volume, 10 times of height to diameter, and 6 BV x h(-1) of flow velocity, at the temperature of 25 degrees C. During the removal process, obvious changes of solid content and HPLC profiles were not observed.</p><p><b>CONCLUSION</b>MPS could be used to remove plumbum ion from extract of L. japonica.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the inhibitory effect of total bufadienolides from toad venom against H22 tumor in mice and preliminarily analyze the structures of the metabolites in tissues.</p><p><b>METHOD</b>HPLC and LC-MS were used for analysis of the chemical composition of TBFs. High, middle and low dosages of TBFs were orally administered or intra-peritoneally injected to H22 tumor-bearing mice for thirteen days. The animals were killed and the tumors were stripped and weighed. The metabolites in the tissues such as heart, liver, spleen, lung and kidney, were analyzed by HPLC and LC-MS.</p><p><b>RESULT</b>The chemical composition of TBFs were identified by comparison of the retention times with those of reference substances, on-line UV spectra and MS data. Its main components are concerned with gamabufotalin, arenobufagin, bufotalin, resibufagin, cinobufotalin, bufalin, cinobufagin and resibufogenin. TBFs had no obvious influence on body weight of H-22 tumor-bearing mice orally administered and the inhibition rate against tumor were 14.76%, 16.38% and 10.32% for low (5 mg x kg(-1)), middle (10 mg x kg(-1)) and high dosage (20 mg x kg(-1)), respectively. The mice intra-peritoneally injected with middle and high-dose of TBFs gained body weight slower than the control mice on the 5th day and recovered on the 13th day. The inhibition rate against tumor were 17.30%, 19.80% and 40.95% for low (1.5 mg x kg(-1)), middle (3 mg x kg(-1)) and high dose (6 mg x kg(-1)), respectively. The inhibitory effect took on dose-dependent manner. Based on the HPLC analyses on heart, liver, spleen, lung and kidney, bufadienolides were found in the liver tissue and 11 compounds of them were tentatively identified by LC-DAD-MS.</p><p><b>CONCLUSION</b>TBFs by oral administration had no inhibitory effect against H22 tumor in mice, however, TBFs by intra-peritoneal injection displayed the significantly inhibitory effect, accompanying some toxicity for early duration of the study. The identification of bufadienolides in the liver provides a good basis for the further investigation of the metabolic pathways of TBFs in vivo.</p>

ABSTRACT

A HPLC method for limit detection of aristolochic acid A in the Chinese herbs containing aristolochic acid or suspected-containing aristolochic acid and their preparations was established. The samples were analyzed on an Alltima C18 column eluted with methanol-water-acetic acid (68:32:1.5) as the mobile phase. Flow rate was at 1.0 mL x min(-1) and the detection wavelength was at 390 nm. The calibration curve was linear over the range from 0.016 to 0.51 g (r = 0.9993) and LOD was 4 ng. The average recovery was 101.2% with RSD of 2.01%. The procedures of sample preparation were systematically investigated. The contents of aristolochic acid A in Radix et Rhizoma Asari bought from market or drugstore were fluctuated from 3.1 to 26.6 microg x g(-1) and 3 of 11 samples accorded with the quality requirement of current Chinese Pharmacopoeia. Among 15 batches samples of Chinese medicaments, only one sample was found to contain aristolochic acid A. The present investigation shows that the method is sensitive and repeatable and it could be used for the limit detection of aristolochic acid A in the Chinese herbal medicines containing trace amount of aristolochic acid A or suspected-containing aristolochic acid A and their preparations.

ABSTRACT

<p><b>OBJECTIVE</b>To develop a quantitative determination of Forsythia suspensa and to provide scientific basis for its quality control.</p><p><b>METHOD</b>By HPLC, the separation was performed on a Zorebax Eclipse XDB-C18 column (4.6 mm x 250 mm, 5 microm) at 25 degrees C using a isocratic elution of acetonitrile-water (containing 0.4% glacial acetic acid, 15:85). The detection wavelength was 330 nm.</p><p><b>RESULT</b>The calibration curve showed a good linearity (r = 0.9998) within test range of 0.202-1.515 microg. And the average recovery was 98.54% with RSD 1.1%. The content range of forsythoside A in 11 batches of Forsythiae Fructus was 0.200%-1.681%.</p><p><b>CONCLUSION</b>The developed method is simple, accurate, specific, and with good repeatability. It is suitable for quality evaluation of Forsythiae Fructus.</p>
